They developed an experiment to determine how DNA replicates. The experiment tested Watson and Crick's model of replication. They distinguished old DNA and synthesized DNA. Labeled them with isotopes, heavy nitrogen-14 and light nitrogen-14, raised E. coli and spun it. They showed that DNA replicates by a semi-conservative mechanism. Double helix separates and each old strand is a template for the new strand.
